Note:The diastereoselective formation of cyclic nucleoside phosphoramidites has been studied. It has been found that six-membered cyclic nucleoside phosphoramidite 13 is a relatively stable intermediate for the synthesis of oligonucleotides. Thermodynamic driving force favors the diastereomer with an axial thymidine substituent in six-membered cyclic phosphoramidites. A group of triazoles and imidazoles have been tested as activators for stereoselective coupling reaction of nucleoside phosphoramidites with alcohols. While some catalysts with stronger acidity lead to more rapid coupling, those with larger sizes gave higher stereoselectivity.
