The toxic oil syndrome: contribution to its ethiology by xenobiochemical studies on 3-(N-phenylamino)-1,2-propanediol

[cat] Diferents estudis epidemiològics han indicat els derivats mono- i diesterificats del 3-(N-fenilamino)1,2-propandiol (PAP) com a biomarcadors de les partides d'oli de colza que van causar la síndrome de l'oli tòxic (SOT), una intoxicació que va tenir lloc a Espanya al 1981, i que va provocar més de 400 morts i va afectar a més de 20,000 persones. Aquests esters del PAP poden ser hidrolitzats per part de la lipasa pancreàtica humana donant lloc a PAP en un últim estadi. Per aquest motiu, els estudis de metabolisme, tant de primera com de segona fase, s'han centrat en aquesta molècula. Així, incubacions de PAP en presència de microsomes hepàtics han mostrat que el CYP450 és capaç de bioactivar aquest compost donant lloc al corresponent derivat 4'-hidroxilat com a principal metabòlit. Aquest fenol derivat del PAP, però, és força inestable i es pot tornar a oxidar donant lloc a una espècie iminoquinònica altament reactiva, i per tant, de potencial toxicitat. En presència de molècules amb un grup tiol lliure, com són el glutatió o l'N-acetilcisteïna, aquesta iminoquinona derivada del PAP pot formar els corresponents tio-conjugats mitjançant una addició de Michael. En aquest cas, els conjugats es poden tornar a oxidar donant lloc als corresponents derivats iminoquinònics, els quals, en presència de glutatió o N-acetilcisteïna formen bis- i tris-adductes, però en absència d'aquests tiols isomeritzen a la corresponent base de Schiff i posteriorment s'hidrolitzen per donar lloc als corresponents conjugats derivats del 4-aminofenol juntament amb gliceraldehid. Aquest comportament es podria extrapolar a d'altres derivats del 4-aminofenol que continguin un CH2 adjacent al residu anilínic. Aquest seria el cas d'un dels possibles metabòlits de la 3-(N-fenilamino)alanina (PAA), un dels contaminants presents en els lots d'L-triptòfan responsables de la síndrome de l'eosinofília-miàlgia (SEM), una altra intoxicació de característiques similars a la SOT que va tenir lloc l'any 1989 als Estats Units i que va afectar a més de 1500 persones. Així, s'ha demostrat que la PAA en presència de microsomes de fetge humà pot ser metabolitzada pel CYP450 donant lloc al corresponent derivat 4'hidroxilat, el qual es pot re-oxidar fàcilment per formar el corresponent intermedi iminoquinònic i donar lloc posteriorment al 4-aminofenol i a la formilglicina. Aquest fet estableix un nou vincle d'unió entre ambdues síndromes, no només a través de la generació d'un compost comú de coneguda toxicitat (4-aminofenol), sinó també mitjançant la generació d'espècies carboníliques que podrien representar un factor de risc addicional.[eng] Toxic Oil Syndrome (TOS) was a massive food intoxication episode that occurred in 1981 in central and northern Spain, causing more than 400 deaths and affecting more than 20,000 people. TOS was attributed to the ingestion of a rapeseed oil that had been adulterated with aniline and illegally refined for human consumption. Epidemilogical investigations established the mono- and diester derivatives of 3-(N-penylamino)-propane-1,2-diol as biomarkers of the toxic oil. These PAP esters can be hydrolyzed by pancreatic lipase leading finally to PAP. For this reason, metabolism studies have been focused in this molecule. Thus, PAP incubations in presence of liver microsomes have shown the capacity of CYP450 to bioactivate this compound giving rise to the 4'-hydroxylated derivative as the main metabolite. However, this phenol derivative is unstable and can be re-oxidated leading a highly reactive, and potentially toxic, quinoneimine specie. In presence of molecules with a free thiol group, as glutathione or N-acetylcysteine, this quinoneimine derivative can forme the corresponding thio-conjugate. However, these conjugates can be re-oxidated giving rise to the corresponding quinenimine derivatives, which in presence of glutathione or N-acetylcysteine lead to bis- and tris-conjugates, but in absence of thiols can isomerize to the corresponding Schiff base and then be hydrolyzed to lead the corresponding 4-aminophenol conjugates and glyceraldehyde. This behaviour could be extrapolated to other species with a CH2 adjacent to the anilinic residue. This could be the case of one of the possible metabolites of 3-(N-phenylamino)-alanine (PAA), one of the contaminants present in the L-tryptophan batches responsible for Eosinophilia-Myalgia Syndrome (EMS), another similar intoxication that took place in 1989 in US and that affected more than 1500 people. Thus, it has been demonstrated that PAA in presence of human liver microsomes can be metabolized by CYP450 to give rise the corresponding 4'-hydroxy derivative, which can be re-oxidated leading the corresponding quinoneimine intermediate and then 4-aminophenol and formyl glycine. This fact provides support of a linkage between EMS and TOS, not only by the generation of a final common toxic compound (4-aminophenol), but also by the concomitant release of carbonyl especies that could represent an additional risk factor
