UJI POTENSI KEMAMPUAN BIOREMIDIASI DAN BIOFERTILIZER BAKTERI LUMPUR AKTIF (SLUDGE) INDUSTRI PT SURABAYA INDUSTRIAL ESTATE RUNGKUT (SIER)

Heavy metal waste produced by industry is classified as B3 waste. Some adapted microbial mechanisms in metal-contaminated environments include microbes capable of using metal as an energy source, precipitating metals in the form of insoluble metal salts, immobilizing metals in cell walls, producing chelating agents, changing the permeability of microbial cell membranes to metals, and reduce the metal to a non-toxic form. This study aims to determine the colony diversity of bacterial types contained in activated sludge, the tolerance level of isolated isolates from activated sludge against environmental stress and heavy metal contamination based on their growth patterns, as well as the ability of isolates of bacteria with the best tolerance level in produce a plant growth hormone and secondary metabolites. This study uses research stages, namely sampling, tool sterilization, manufacture of solid LB media, bacterial isolation, isolation of isolates, stock-culture, gram staining, re-culture isolates, and GC-MS analysis. Bacterial isolation carried out resulted in 14 bacterial isolates by purifying bacteria with morphological diversity of cells and varying cell colony morphology. Heavy metal tolerance test was carried out on 6 bacterial isolates namely S1aB1, S1aB2, S1bB4, S2aB5, S3aB7, and S4bB11. S4bB11 isolates become bacteria that have the best growth in environmental stress and heavy metal stress. The results of GC-MS analysis on S4bB11 bacterial isolates, it is known that metabolites that play a role in the bioremediation process as a form of adaptation to heavy metal stress environments, namely α ketoglutaric acid, diaminopimelic acid, and mannose. 3 indoleacetic acid and kinetin are the results that can act as biofertilizers for plant
