Investigation of the basal and cholesterol-dependent trafficking of P-glycoprotein

A humán multidrog rezisztencia transzporter, P-glikoprotein megtalálható a plazmamembrán raft és nem-raft doménjeiben is. A Pgp azon konformációs állapota (Pool1), mely az UIC2 antitesttel azonosítható nagyobb mértékű raft asszociációt mutat, fokozott internalizációval rendelkezik és recirkulál a sejtfelszínre. Ennek eredményeként a Pgp-t expresszáló multidrog rezisztens sejtek jobb életképeségeket mutattak a plazmamembrán koleszterin modulációját követően.The multidrug resistance transporter human P-glycoprotein (Pgp) resides in both raft and non-raft domains of the plasma membrane. The conformer of Pgp that binds the monoclonal antibody UIC2 (Pool1) is primarily associated with raft domains, exhibits an increased internalization and recycle to the cell surface. Depletion of cholesterol by cyclodextrin led to an unexpected increase of the raft-associated fraction of Pgp in the cell membrane as a result of an increased exocytosis. The exocytotic event turned out to be part of a complex membrane repair process initiated by lysosomal exocytosis after cholesterol depletion. The Pgp overexpressing 3T3-MDR1 cells had higher total cell cholesterol, more numerous and stable lysosomes, and they exhibited more effective membrane repair after cholesterol modulations compared to the parental cell line. As a result of these, the multidrug resistant Pgp-overexpressing tumor cells exhibited significantly better viabilities after cholesterol modifications.d
