Molecular taxonomy of Crocus vernus (Iridaceae) species complex in the Carpathian Basin

A Crocus vernus fajcsoport (Iridaceae) morfológiailag és citogenetikailag változatos növénycsoport, amelynek nevezéktana rendkívül bonyolult. Kutatómunkám a C. vernus fajcsoport 5 Kárpát-medencei taxonjának filogenetikai vizsgálatára irányult. A genetikai variabilitás vizsgálata a plasztiszban kódolt accD-psaI intergénikus régió szekvenciájának vizsgálatával és amplifikált fragmenthossz polimorfizmus (AFLP) alapján történt. Az AFLP és plasztid filogenetikai fa a vizsgált taxonokat két csoportra osztotta: (1) adriai és (2) balkáni kládokra. A balkáni csoportot a C. heuffelianus populációk, valamint a C. tommasinianus egyedei alkotta. Az adriai kládhoz C. vittatus és C. vernus populációk, valamint a C. neapolitanus tartoztak. A C. scepusiensis estén inkongruens eredményt kaptunk, amely a plasztid adatok alapján a balkáni kládba, míg az AFLP adatok alapján az adriai kládba tartozott. A Bayesian analysis a C. scepusiensis-t F1 hibrid genotípusba sorolta, míg a C. vernus és C. heuffelianus taxonokat szülői genotípusba. Kariológiai vizsgálataink eredményei alapján, amely során a C. scepusiensis esetén 2n=18 tetraploid kromoszóma számot állapítottunk meg, azt feltételezzük, hogy a C. scepusiensis faj a balkáni klád egy 2n=10 kromoszóma számú (pl.: C. heuffelianus Erdélyből) tagjának és az adriai klád egy 2n=8 kromoszóma számú (pl.: C. vernus) tagjának allopoliploid hibridje. Hasonló hibrid eredetet feltételezünk a C. vittatus (2n=18) esetén is, amely a Bayesian analízisben szintén F1 hibrid genotípusba tartozott. Eredményeink alapján a Kárpát-medence északi és déli régiójában egy paralell fajkeletkezési folyamat játszódhatott le ugyanazon szülőfajokból kiindulva, amely különböző allopoliploid leszármazottat eredményezett.The Crocus vernus Hill. has an impressive variation in morphology and has various cytological characters and has complicated taxonomic history. My study focused on the investigation of the phylogenetic affinities of five taxa in the C. vernus species complex from the Carpathian Basin. Genetic variability was evaluated using chloroplast DNA sequences of the accD-psaI intergenic spacer and amplified fragment length polymorphism (AFLP). Both the AFLP and plastid phylogenetic trees separated the taxa into two groups: (1) a ‘Balkan’ clade, and; (2) an ‘Adriatic’ clade. The Balkan clade contained C. heuffelianus and as well as C. tommasinianus samples; the Adriatic clade included C. vittatus and C. vernus samples as well as C. neapolitanus. A hard incongruence was found in the placement of C. scepusiensis which clustered inside the Balkan clade on our plastid tree, while it fell in the Adriatic clade on the AFLP tree. The Bayesian analysis assigned all C. scepusiensis specimens as F1 hybrids, while C. vernus and C. heuffelianus were assigned as parental species. Together with our cytological investigation that determined a 2n=18 chromosome number of C. scepusiensis, we conclude an allopolyploid hybrid origin for this taxon as the result of a cross between members of the 2n=10 Balkan clade (e.g., C. heuffelianus from Transylvania) and the 2n=8 Adriatic clade (C. vernus). A similar origin is postulated for C. vittatus (2n=18), which was clustered as an F1 hybrid in the Bayesian analysis; thus, parallel evolution may have taken place in the northern and southern part of the Carpathian Basin beginning from the same parental species but leading to different allopolyploid derivatives.N
