Cross-dependencies between different genes for the ability to confer resistance to HAP when overexpressed.

<p>Each plasmid was transformed into yeast knockout strains where one of the other resistance genes had been deleted. Overexpression plasmids (see <a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0196840#pone.0196840.g001" target="_blank">Fig 1</a>) are shown at the top, yeast strains at the left, and drug concentrations at the bottom in each subfigure. Transformants were grown in liquid medium to late exponential phase, diluted, and then spotted onto uracil- and adenine-less SC galactose (Gal-UA) plates with or without 300 μg/ml HAP. Note that the <i>ade4</i>, <i>ade1</i>, and <i>ade2</i> strains do not grow on adenine-less media in the absence of the drug, but are able to grow weakly in the presence of HAP which is partially deaminated to adenine. The + sign stands for the haploid wild type control strain BY4742, +/+ stands for the diploid wild type control strain BY4743, and +/<i>dut1</i> for the diploid <i>DUT1/dut1</i> heterozygote. The control vector is pHR81 [<a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0196840#pone.0196840.ref030" target="_blank">30</a>]. The no drug control plate was photographed after 3 days and the HAP plate after 5 days.
