<p>The sgRNA barcode in Perturb-seq is part of the puromycin resistance gene / BFP transcript which is driven by core EF1α promoter (upper panel). Mosaic-seq and CRISP-seq share a similar design, in which the barcode is inserted immediately upstream of the lentiviral 3’LTR (middle panel). In CROP-seq, the sgRNA-expressing cassette is inserted into the 3’LTR, allowing direct detection of sgRNA sequences.</p
