An In Vitro screening of bile salt hydrolase activity in probiotic Lactobacillus strains

Kardiovaskulära sjukdomar är den främsta dödsorsaken i världen och orsakas främst av ateroskleros. Den nuvarande behandlingen av ateroskleros utgörs främst av förbättrad kost, träning och konventionell behandling med kolesterolsänkande läkemedel. De har vissa begränsningar och det finns behov av ytterligare alternativ och kompletterande behandling. Probiotiska Lactobacillus stammar som producerar gallsalthydrolas har uppvisat kolesterolsänkande egenskaper i kliniska studier. Det har därför förekommit mycket forskning inom detta ämne för att kunna identifiera dessa stammar som vidare kan användas för kardiovaskulär behandling. Syftet med denna studie var att undersöka gallsalthydrolas aktiviteten i Lactobacillus stammar. Utvalda Lactobacillus stammar screenades för gallsalthydrolas aktivitet gentemot de två gallsalterna glycodeoxycholat syra (GDCA) och taurodeoxycholat syra (TDCA). Mätningar genomfördes med en kvalitativ plattmetod och en kvantitativ reversed phase-high performance liquid chromatography (HPLC) metod, där dekonjugering av gallsalterna analyserades. Även spektrofotometriska mätningar gjordes för att undersöka Lactobacillus stammarnas tillväxt under inverkan av gallsalterna. Resultaten visade på att bakteriestammarna hade gallsalthydrolas aktivitet som var substratspecifika och reagerade olika gentemot GDCA och TDCA. De uppvisade även en högre tendens till att dekonjugera GDCA än TDCA. En tydlig tillväxtberoende reglering av gallsalthydrolas aktiviteten kunde observeras. Alla Lactobacillus stammar utom en uppvisade aktivitet av gallsalthydrolas och kan därmed vara potentiella kandidater för behandling av kardiovaskulära sjukdomar. Vidare in vitro studier behövs för att påvisa resultatens reproducerbarhet och även in vivo studier för att utvärdera de probiotiska Lactobacillus stammarnas inverkan på den kardiovaskulära hälsan.Cardiovascular diseases (CVD) are the leading cause of global mortality where atherosclerosis is the leading causative factor. Current treatments for atherosclerosis include diet changes, exercise, and conventional cholesterol-lowering drugs, however they have limitations and additional treatments are needed. Probiotic bile salt hydrolase (BSH) active bacteria such as Lactobacillus strains have demonstrated cholesterol lowering abilities in clinical trials, therefore much research has been focused on identifying strains that could be used in CVD treatment. The main aim of this project was to investigate BSH activity in Lactobacillus species. In this study selected Lactobacillus strains were screened for BSH activity against two bile salts, glycodexycholic acid (GDCA) and taurodeoxycholic acid (TDCA) using the qualitative direct plate assay and quantitative analysis of deconjugation by a reversed phase-high performance liquid chromatography (RP-HPLC). The influence of bile salts on growth of the Lactobacillus strains was measured spectrophotometrically. The results showed that the strains have a BSH activity dependent on the glycine or taurine amino acid moiety and a higher tendency to deconjugate GDCA compared to TDCA. BSH activities are also growth phase regulated. All the strains except one showed BSH activities and they could have potential for probiotic CVD treatment through cholesterol lowering abilities. Further in vitro studies must be performed on the Lactobacillus strains to confirm the reproducibility of the results, and in vivo studies are needed to evaluate their effect on the cardiovascular system
