Transmission electron microscopy in cell biology: assessment of techniques and image information

La microscòpia electrònica de transmissió dins l’àmbit de la biologia ceŀlular és una tècnica consolidada i una eina molt útil en la recerca biomèdica. Dins de les microscòpies és la que té un límit de resolució més alt i, per tant, la fa una tècnica molt eficient per interpretar l’arquitectura ceŀlular i poder relacionar aquesta estructura amb la seva funció. La innovació i la millora d’equipaments, juntament amb el desenvolupament de noves tecnologies, permeten assolir un millor coneixement del teixit biològic, una millor visualització de les estructures i la identificació i localització de molècules. Es defineixen processos i conceptes, valorant els avantatges i desavantatges de cada tècnica, la imatge i la informació final que podem obtenir utilitzant cadascuna.Paraules clau: microscòpia electrònica de transmissió, fixació química, criofixació, noves tecnologies, informació de la imatge.Transmission electron microscopy is a proven technique in the field of cell biology and a very useful tool in biomedical research. Of all the types of microscopy used to date, transmission electron microscopy is the one with the best resolution limit and therefore it is a very efficient technique for deciphering cell architecture and relating it to function. Innovation and improvements in equipment together with the introduction of new technology have allowed us to improve our knowledge of biological tissues, to visualize structures better, and to identify and locate molecules. In this paper, processes and concepts are defined, and the advantages and disadvantages of each technique are assessed, describing the image and information that can be obtained with each one.Keywords: transmission electron microscopy, chemical fixation, cryofixation, new technologies, image information
