Somatic cells can be reprogrammed into induced pluripotent stem (iPS) cells by overexpression of combinations of transcription factors such as Oct4, Sox2, Klf4 and c-Myc. However, reprogramming is slow and stochastic, suggesting the existence of mechanisms that limit its efficiency. Senescence is an irreversible G1 cell cycle arrest elicited by replicative exhaustion or in response to stresses such as DNA damage, or aberrant expression of oncogenes. The arrest observed during senescence is implemented mainly through activation of p53 and the upregulation of the cyclin-dependent kinase (CDK) inhibitors, p16INK4a and p21Cip1.
