Rearrangements involving the mixed lineage leukemia gene (MLL) are found in majority of human infant leukemias (>60% of ALLs, 35% of AMLs) and are associated with dismal prognosis of these patients. They are also found in a small percentage of childhood and adult leukemias (10% cases), making MLL-rearranged leukemias ideal for this study. We performed induction of the leukemic MLL-ENL chromosomal translocation in hematopoietic cells at different developmental stages i.e., fetal liver (FL, E12.5) and bone marrow (BM, P60) to develop disease models that can recapitulate human infant and adult leukemias respectively.
