Yeast transcriptional activator GCN4(226-252)-GGC-NH2 and the designed Ac-[Q(236), Q(242)]GCN4(226-252)-GGC-NH2 aiming at altering GCN4 DNA-binding specificity was prepared by stepwise solid phase peptide synthesis, It was found that the yield of the peptide would be greatly improved when treated with dithiothreitol (DTT).
