目的:观察三氧化二砷(As2O3)对非急性早幼粒细胞性白血病(acute prom y elocytic leukemia, APL)急性髓性白血病细胞的体外作用以及维甲酸(retinoic acid, R A )与As2O3之间的相互作用.方法:用维甲酸敏感(HL-60S)及维甲酸耐药(HL-60R)急性髓性白血病细胞株HL-60作为体系, 应用细胞计数及台盼蓝试验观察细胞活力及生长.采用细胞形态、DNA 片段梯度、末端核苷酸酶标记及流式细胞法观测细胞凋亡.结果: As2O3(0.5~4 μmol·L-1)抑制 HL -60R细胞生长,抑制能力与As2O3浓度及作用时间呈正相关;RA不仅促进HL-60S而且促进HL-60R 对As2O3的敏感性.结论:As2O3对细胞的抑制作用不局限于PML/RARα阳性细胞;As2O3可能对非APL急性髓性白血病有治疗作用 ,RA 与As2O3合用可能发挥更佳效果.中国科学引文数据库(CSCD)06552-5553
