采用微团培养观察了不同浓度的双酚A(0~50mg/L)对体外培养的Wistar大鼠胚胎肢芽细胞增殖和分化的影响,旨在了解双酚A的体外发育毒性特点及其可能的作用机制.结果显示:双酚A对体外培养的Wistar大鼠胚胎肢芽细胞的增殖和分化均有抑制作用,表现为染毒组肢芽细胞集落形成数量减少,细胞毒性试验吸光度值下降,并呈现出明显的剂量-反应关系.双酚A对肢芽细胞的50%增殖抑制浓度(IP50)和50%分化抑制浓度(ID50)分别为38.74mg/L 和27.93mg/L,IP50/ID50=1.39.按照Renault等人推荐的体外致畸分类标准,双酚A属于阳性致畸物,对肢芽细胞的分化和增殖有特异性抑制作用.The micromass culture of Wistar rat embryo limb bud cells was used to investigate the characteristic of developmental toxicity of Bisphenol A (BPA) and its mechanism in vitro.
