This thesis describes the design of several tool for mechanical stimulation at cellular scale, and their use for muscle cell rheology and sutdy of the MRTF/SRF signalisation pathway. Exerting and feeling forces is a main role of muscle cells. Their developement depends on mechanical stimulation : physical training leads to growth of muscles whereas disuse leads to atrophy. The actin/MRTF/SRF pathway is central in the mechanotransduction, by connecting the mechanosensitive actin cytoskeleton with transcription factors that control muscle developpement gene expression.
