The aim of this work was the study of intracellular factors responsible for the macroheterogeneity of recombinant human IFN-gamma (IFN) glycosylation produced in bioreactor during batch and fed-batch cultures of Chinese Hamster Ovary (CHO) cells. Analysis of oligosaccharide precursors revealed that the structure of the lipid-linked oligosaccharides was constant over the batch process. On the contrary, the hypothesis of a quantitative limitation of the glycan structures could not be excluded. Moreover, the decline in intracellular nucleotide triphosphates and adenylate energy charge could be linked to the decrease in IFN glycosylation.
