Effect of <i>PRKD3</i> gene silencing on the expression of AP-1 factors in human chondrocytes.

<p>Following transfection with siRNA specific to <i>PRKD3</i> (black bars) or non-targeting siCon (white bars) (both 100 nM), primary human articular chondrocytes were stimulated with IL-1+OSM as detailed in the Methods for 1 h unless stated otherwise. (A) Cell lysates were subjected to real-time RT-PCR (n = 6; mean ± S.E.) for <i>FOS</i> (<i>upper panel</i>) or <i>JUN</i> (<i>lower panel</i>) as described in Methods. Data are presented as relative expression levels normalised to <i>18S rRNA</i> housekeeping gene, where **, p≤0.01 versus siCon. (B and C) Nuclear fractions were prepared and subjected to SDS-PAGE and immunoblotting using the antibodies indicated. Scans from multiple blots were combined and the relative density values presented for the individual antibodies used (C), where **, p≤0.01 vs the relevant siCon time point.
