EF-1A levels commensurately affect both <i>trm7Δ</i> growth and activation of the GAAC response.

<p><b>(<i>A</i>) An additional copy of EF-1A partially suppress the growth defect of <i>trm7Δ</i> mutants.</b><i>trm7Δ</i> mutants containing a [<i>CEN LEU2</i>] plasmid with <i>TEF1</i>, <i>TEF2</i>, or <i>TRM7</i> as indicated were grown overnight in SD-Leu at 30°C, analyzed by spotting to plates as indicated, and incubated for 2 d at 30°C. <b>(<i>B</i>) An additional copy of EF-1A partially suppresses induction of the GAAC response.</b> WT or <i>trm7Δ</i> strains containing a [<i>CEN LEU2</i>] plasmid with <i>TEF1</i> or <i>TEF2</i> as indicated were grown as in (<i>A</i>), and relative <i>HIS5</i> mRNA levels were determined. p values are shown above. <b>(<i>C</i>) Reduced levels of EF-1A amplify the growth defect of <i>trm7Δ</i> mutants.</b> WT and <i>trm7Δ</i> [<i>CEN URA3 TRM7</i>] strains with or without a <i>tef2Δ</i> mutation, as indicated, were grown overnight in rich media, and analyzed by spotting to media containing 5-FOA, and incubation for 2 d at 30°C. Strains from the 5-FOA plate were then purified on medium containing 5-FOA, grown overnight in YPD, diluted, spotted on rich (YPD) and minimal (SD complete) plates as indicated, and incubated for 2 d at indicated temperatures.
