Macrophage migration towards <i>C. albicans</i>.

<p>Differences in macrophage velocity were observed when macrophages were cultured with different <i>C. albicans</i> glycosylation mutants. Tracking software was used to conduct a detailed dissection of macrophage migration dynamics. <a href="http://www.plospathogens.org/article/info:doi/10.1371/journal.ppat.1002578#ppat-1002578-g001" target="_blank">Figures 1A, B and C</a> are snapshots from a tracking movie showing an individual macrophage (red, *) in pursuit of live wildtype <i>C. albicans</i> (green). Initially, the macrophage's movement appeared to be random (1A). After the macrophage had sensed <i>C. albicans</i>, it accelerated and homed in on its target (1B) leading to engulfment (1C). <a href="http://www.plospathogens.org/article/info:doi/10.1371/journal.ppat.1002578#ppat-1002578-g001" target="_blank">Figures 1D and E</a> show tracking diagrams illustrating the distances travelled, directionality and velocity of J774.1 macrophages cultured with live wildtype and <i>mnt1</i>Δ<i>mnt2</i>Δ glycosylation mutant, respectively. Due to the large number of macrophages tracked per video, the data was filtered to show only active macrophages with a mean track velocity greater than 1.80 µm/min. Tracks represent the movement of individual macrophages relative to their starting position, symbols indicate the location of macrophages at 1 min intervals and arrows represent directionality.
