Knockdown of AP-1 subunits decreases Blimp1 expression in lung cancer cells.

<p>(A) The immunoblot of nuclear extracts from lung cancer cells in <a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0033287#pone-0033287-g001" target="_blank">Fig. 1A</a> was stripped and re-probed to assess expression of the AP-1 subunits c-Jun, Fra-1, Fra-2 and c-Fos. (B) A549, H441 and H1299 cells were transfected with 20 nM of <i>JUN</i> siRNA alone or 10 nM of <i>JUN</i> siRNA in combination with 10 nM of <i>FRA-1</i> or <i>FRA-2</i> siRNA or with 20 nM of a negative control siRNA (Qiagen) for 24 h. WCE (30 µg) were subjected to immunoblotting for Blimp1, c-Jun, Fra-1, Fra-2 and α-tubulin, as a loading control. The Blimp1 bands were quantified and normalized to α-tubulin expression, and average values from two independent experiments presented relative to control siRNA, set to 1.0.
