Preparation of DH01 cells under transplant conditions does not induce cell death.

<p>(<b>A</b>) When 20×10⁶ cells (20) were seeded they exhibited significantly (p<0.01) higher levels of nonviable cells than flasks seeded with 0.5×10⁶ cells (0.5) as measured by trypan blue exclusion, however, transplant conditions (TC) did not significantly affect viability compared to baseline conditions (BC) (p = 0.799). Data are given as mean±SD and are representative of 3 independent experiments. (<b>B</b>) SDS-PAGE analysis of caspase 3 and 7 expression in cells seeded at varying cell densities (0.5×10⁶ cells and 20×10⁶) and prepared under baseline and transplant conditions using antibodies sensitive for the pro- and cleaved forms. There was no change in the expression of either caspase 3 or 7 (pro-form) and we did not detect the cleaved fragments.
