Knockdown of DcR3 inhibits TRAIL-induced cFLIP_L upreguation and NF-κB activation in HepG2 cells.

<p><b>(A)</b> In the presence or absence of shDcR3 and z-VAD, HepG2 cells were treated with TRAIL (75ng/ml) for 24 h. Western blot was used to detect the protein level of cFLIP_L. (B) Real time PCR was used to detect the mRNA level of cFLIP_L. (C) After infection with shDcR3 for 12 h in the presence or absence of 100 μ M z-VAD-fmk, HepG2 cells were treated with TRAIL (75ng/ml) for 24 h. The levels of cytosolic and nuclear p65 proteins were determined by western blots. (D) Luciferase activity was determined by a luminometer in cell lysates.
