Knockdown of DcR3 inhibits TRAIL-induced NF-κB activation.

<p>After infection with shDcR3 for 12 h in the presence of 100 μ M z-VAD-fmk, Huh7 cells were treated with TRAIL (75ng/ml) for 24 h. (A) The levels of cytosolic and nuclear p65 proteins were determined by western blots. (B) The nuclear translocation of NF-κB p65 were examined by confocal microscopy. Original magnification ×400. (C) Luciferase activity was determined by a luminometer in cell lysates. (D) Western blot was used to determined the IKK α / β phosphorylation and I κ B phosphorylation/degradation.
