Les cassures double-brin (CDBs) de l’ADN sont généralement néfastes pour la cellule, toutefois il existe une exception importante à cette règle. Chez la levure et les autres eucaryotes, Spo11 (ou Rec12 chez S. pombe) est décrit comme étant capable de créer des CDBs, bris de l’ADN nécessaire à l’initiation de la recombinaison méiotique. Des analyses génétiques et bioinformatiques ont démontré que les homologues de Spo11 lient l'ADN double-brin sous la forme d’un dimère.
