Acyclovir is a synthetic purine nucleoside analogue with in vitro and in vivo inhibitory activity against herpes simplex virus types 1 (HSV-1), 2 (HSV-2), and varicella-zoster virus (VZV). The efficacy of oral acyclovir is limited as a result of its low bioavailability (15-30%) as it is poorly water soluble and therefore requires a frequent dosing regimen. When orally administered, peak plasma concentration occurs after 1.5–2.5 hours, while its elimination half-life is approximately 2-3 hours. Acyclovir displays poor solubility in water and in lipid bilayers, which leads to poor drug levels at target sites after oral, local, or parenteral administration.
