<p>(A) The depletion of DDRGK1 decreases NF-κB activity. U2OS cells were transfected with either mixed DDRGK1 siRNAs or a control siRNA, along with the control pGL6-TA or the pNF-κB luciferase reporter. 48 hours after transfection, the cells were treated with 10 ng/ml TNF-α for 4 hours or left untreated as control. Luciferase activities were determined by the dual luciferase assay system. (B) The depletion of DDRGK1 inhibits the DNA-binding activity of p65. U2OS cells were transiently transfected with mixed DDRGK1 siRNAs or a control siRNA, and nuclear proteins were extracted. The DNA-binding activity of p65 toward a consensus κB site was then assessed using the TransAM NF-κB p65 transcription factor assay kit.</p
