The mAb D6F10 binds and internalizes in HeLa cells along with abrin.

<p>0.04 million HeLa cells adhered on a cover slip were incubated with 1∶50 dilution of normal goat serum in complete DMEM for 1 h at 37°C or 4°C with 0.01% azide. Cells were then incubated for 45 min with 100 ng abrin and 10 fold molar excess of the antibody, washed twice with PBS with/without lactose and fixed with paraformaldehyde. After two washes with PBS, cells were incubated with 1∶500 dilution of anti-mouse Alexa-633 labelled secondary antibody for 1 h at room temperature. Following this, the cells were stained for their nuclei with propidium iodide (represented as blue color) containing RNase, washed again and mounted on slides. The stained cells were observed under a Zeiss confocal scanning microscope and Z-stacks separated by 0.75 µm were imaged.
