Blockage of p38MAPK signaling decreased the expression of IL-1α in primary PDAC cells.

<p>(<b>A–E</b>) The primary PDAC cell line PC013 was cultured for 24 h in 1% FBS medium supplemented with vehicle (DMSO) or different concentrations (0–150 µM) of the p38MAPK inhibitor SB203580, the ERK inhibitor U0126 (0–50 µM), or the JNK inhibitor SP600125 (0–75 µM). The mRNA expression levels of (<b>A–C</b>) IL-1α, (<b>D</b>) IL-1β, and (<b>E</b>) IL-1RA were analyzed using qRT-PCR. (<b>F</b>) PC013 cells were lysed after 24 h, 48 h, and 72 h after incubation with 100 µM SB203580 or vehicle and IL-1α was measured by ELISA and the result presented in pg/ml per 1×10⁵ cells.
