The role of TNF and caspases on rJHP0290-induced macrophage apoptosis.

<p>(<b>A</b>) RAW264.7 cells were pretreated either with isotype control IgG or TNF neutralizing antibody (10 µg/ml) for 1 h followed by treatment with either rJHP0290 (2 µg/ml) or LPS (500 ng/ml) for 24 h. Apoptotic cells were analyzed by staining with AnnexinV antibody and PI using Flow cytometry. (<b>B</b>) RAW264.7 cells were treated either with DMSO or caspase inhibitor Z-VAD-FMK (20 µM) for 1 h followed by treatment with rJHP0290 (2 µg/ml) for 24 h. Percentage of apoptotic cells was determined. Values indicate mean ± SD of three independent experiments. Statistically significant differences are indicated by *(p<0.05).
