The R39C <i>unc-18</i> point mutation suppresses the <i>rab-3</i>-dependent resistance to aldicarb.

<p><i>Loss-of-function </i>(<i>lof</i>) <i>rab-3</i> (<i>y250</i>) worms were created to express transgenically wild-type (Wt) <i>unc-18</i> or <i>unc-18</i> with the indicated mutations. In comparison to Bristol N2 worms, <i>lof </i><i>rab-3</i> worms exhibited resistance to aldicarb. This resistance to aldicarb was blocked partially by expression of R39C <i>unc-18</i>. The R39C/E465K double mutation did not affect <i>rab-3</i> aldicarb sensitivity. Significance was assessed by two-way analysis of variance with post-hoc comparisons. Only worms expressing R39C (P<0.001) had altered aldicarb sensitivity in comparison to <i>rab-3</i> worms. N = 3 experiments per strain (20-25 worms each experiment).
