T-loop phosphorylation of Akt at Thr³⁰⁸ in Raptor or PDK1 immunoprecipitates of 1-LN or DU-145 cells transfected with dsPDK1 or dsRaptor-RNAi, and stimulated with α₂M*.

<p>The T-loop phosphorylation was determined by kinase assay. The bars in the diagram are: (1) lipofectamine + buffer; (2) lipofectamine + α₂M* (50 pM/20 min); (3) lipofectamine + dsRaptor RNAi (100 nM/48 h) or dsPDK1 RNAi (100 nM/48 h) then α₂M*; and (4) scrambled dsRNA (100 nM/48 h) then α₂M*. The Akt phosphorylation is expresed as fmol [³³P]-γ-ATP incorporated/mg cell protein and is mean ± SE from three to four independent expermients in each case.
