Involvement of c-Src, NADPH oxidase, and PI3K in epidermal growth factor (EGF)-induced Akt Ser473 phosphorylation in HT-29 cells.

<p>HT-29 cells were transiently transfected with 0.5 µg of pcDNA or 0.5 µg of a dominant negative mutant of c-Src (c-Src DN) (A) for 24 h, or cells were pretreated for 30 min with 10 µM DPI (B) or 10 µM LY 294002 (C) and then stimulated with 10 ng/ml EGF. After 20 min of stimulation, Akt Ser473 phosphorylation was determined. Immunoblots are representative of three experiments with similar results.
