VA RNAI and VA RNAII processing in HAd infections.

<p>HeLa cells were infected with the indicated viruses. 24 hpi total RNA was extracted and analyzed by Northern blot. VA RNAI accumulation and processing into mivaRNAI was analyzed using ³²P-labelled probes complementary to the VA RNAI 5′ stem (<b>B</b>) or the VA-RNAI 3′ stem (<b>C</b>). VA RNAII accumulation and processing into mivaRNAII was detected using ³²P-labelled probes complementary to the VA RNAII 5′ stem (<b>E</b>) or the VA RNAII 3′ stem (<b>F</b>). The detection of tRNA-lysine served as a loading control. Horizontal black bars on the schematic VA RNAI and VA RNAII drawing indicate the localization of the Northern blot probes (<a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0105746#pone.0105746.s007" target="_blank">Table S2</a>). The experimentally verified secondary structures of VA RNAI (<b>A</b>) and VA RNAII (<b>D</b>) are indicated. The different panels were repeated at least two times.
