Overexpression of CDKN3 significantly reduces the efficiency of Bcr-Abl-mediated FDCP1 cell transformation.

<p>(A) Shown are constructs of bicistronic retroviruses carrying Bcr-Abl and GFP, Bcr-Abl and CDKN3-Flag, or empty vector (EV). (B) 293T cells were transfected with the plasmids described in (A). Cells were harvested after 36 hours and protein expression was detected by Western blotting using indicated antibodies. (C) FDCP1 cells were infected with retroviruses carrying empty vector, Bcr-Abl and GFP, or Bcr-Abl and CDKN3-Flag, and plated on 96-well plates. Transformation efficiency was assessed as described in <a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0111611#s2" target="_blank">Materials and Methods</a>.
