I4R-9r variant PwSNs mediated gene silencing <i>in vitro</i> and <i>in vivo</i>.

<p>(A) The relative amount of GAPDH knockdown was determined by Western blot. The intensities of immunoblot was normalized with those of beta actin control and results are presented as mean ± standard deviation (n = 5, (α) ANOVA test, P<0.05: (_<b>**</b>) t-test, P<0.01). (B) To evaluate <i>in vivo</i> gene silencing efficacy, The HT29-luc bearing mice were treated with I4R-9r/siControl, I4R-9r/siLuciferase and I4R-9r(A1*)/sHGP-9r/siLuciferase PwSNs via intra-tumoral injection. Bio-luminescence signals of subcutaneous tumor were monitored using the IVIS Spectrum imaging system in day 0 and 4. (C) The bioluminescence intensities at tumor site were obtained and the relative intensities were calculated based on the bioluminescence intensities of each group in day 0 as 1.
