Wild type RpsL restricts the replication of <i>L</i>. <i>pneumophila</i> strains in macrophages.

<p><b>A</b>. Strain Philadelphia 1 was unable to replicate in bone marrow-derived macrophages. <b>B</b>. Conversion of the <i>rpsL</i>_<i>K88R</i> allele of the laboratory strain Lp02_rpsLK88R into wild type abolished its ability to grow in macrophages. Experiments were performed in macrophages from A/J mice as described in <a href="http://www.plospathogens.org/article/info:doi/10.1371/journal.ppat.1004704#ppat.1004704.g001" target="_blank">Fig 1</a>. With the exception of strains Lp02_rpsLK88R and Lp03, which harbor a spontaneous K88R mutation in <i>rpsL</i>, the <i>rpsL</i> locus of all other bacterial strains was introduced by genetic manipulation. Infections were performed in triplicate and data shown were from one representative of three experiments with similar results.
