Reaction time course of ADAMTS13 and wtVWF73 M13 phage.

<p><i>A</i>, Schematic representation of AlphaLISA reaction in mutant VWF73 bearing phage. Cleavage of the displayed VWF73 peptide was determined by a loss of Alpha signal caused by the release of the FLAG tagged NH₂-terminal VWF73 fragment and subsequent separation of donor and acceptor beads. <i>B</i>, M13 phage displaying wtVWF73 (6 pM) were reacted with ADAMTS13 (2 nM) in ADAMTS13 reaction buffer B at 37C. Aliquots were removed into stop buffer containing EDTA at indicated time points. Data were fit by a first-order non-linear regression model and plotted in the Prism software package.
