<p>(A) HeLa cell cultures were either mock transfected or transfected with identical amounts of cDNAs encoding WT ubiquilin-2 or each of the five different ALS mutations shown. Equal amounts of protein lysate from the transfections, were resolved by SDS-PAGE, transferred to PVDF membrane, and immunoblotted with HA or actin antibodies. (B) Similar to A, but this time the lysates were also probed for S5a and the alpha 6-subunits of the proteasome. (C) One-way ANOVA analysis revealed no statistical change in the levels of the S5a and the alpha 6-subunit in cells transfected with the ubiquilin-2 constructs compared to that in control mock transfected cells.
