Effects of lycopene on H/R-induced loss of cell viability and ER stress.

<p>Primary cultured neonatal mouse cardiomyocytes were pre-incubation with or without 5μM lycopene for 4h prior to 4h of exposure to hypoxia condition with serum-free and glucose-free medium, followed by 1, 2, 4, 6, 8 or 12h of reoxygenation in normal medium. Cell viability was determined by CCK-8 assay. The total mRNA of cardiomyocytes was extracted, and the mRNA levels of GRP78 and GADD153 were detected by Real-time PCR. (C) Time-dependent effect of lycopene on GADD153 mRNA expression in H/R-treated cardiomyocytes. (D) Lycopene reversed H/R-evoked loss of cell viability. Cells were pretreated with or without 5μM lycopene for 4h prior to H/R treatment. Results were presented as a percentage of the control cell survival (set to 100%).
