An analysis for the expression of detoxifying and cytoprotective proteins in the thioacetamide-damaged liver of the rat

Die zugrundeliegenden molekularen und zellulären Veränderungen in der Entstehungsphase des intrahepatischen Cholangiokarzinoms (iCC) sind bisher nur teilweise verstanden. Diverse Mutationen (APC, DCC, OGG1, p53, k-ras, BRAF, TGFßR1, SMAD4) und eine Beteiligung verschiedener Signalwege (PI3K/Akt-, EGFR-, TGF-ß1-, Hippo-, Hedgehog-, Notch-, Wnt-Signalweg) wurden bislang in Bezug auf das iCC beschrieben. Hierbei ist noch nicht geklärt, ob dem iCC möglicherweise eine Adenom-Karzinom-Sequenz zugrunde liegt bzw. zu welchem Zeitpunkt welche Signalwege auf Proteinebene gestört sind. Ziel dieses Projekts war eine Analyse des Proteoms und der Phosphorylierung des Proteoms der Leber während der durch das wasserlösliche Hepatotoxin Thioacetamid (TAA) induzierten Cholangiokarzinogenese. Hierfür wurde ein Rattenmodell mit männlichen Sprague-Dawley-Ratten etabliert, welche ausschließlich Trinkwasser erhielten, dem TAA zugegeben war, und somit dem TAA chronisch ausgesetzt waren. Parallel dazu wurde eine Gruppe mit Kontrolltieren gehalten, welche nur sauberes Trinkwasser erhielt. Die Tiere wurden nach 4, 8, 12, 16 und 18 Wochen geopfert und entsprechend der Leberschädigung histologisch in die Gruppen Fibrose, beginnende Zirrhose, Zirrhose und CC eingeteilt. Die Analyse der Veränderung der Proteinmengen und des Phosphorylierungsstatus wurde mit Hilfe von 2DE-Gelen und anschließender Massenspektrometrie durchgeführt. Hierbei wurden im Vergleich zur Kontrollgruppe bei den behandelten Tieren insgesamt 38 Proteine mit einer mindestens 1,5-fach veränderten Abundanz und 25 Proteine mit einer mindestens 1,5-fachen Veränderung ihres Phosphorylierungsstatus identifiziert. Die Proteine wurden nach Funktionsgruppen sortiert. Nach den am Metabolismus beteiligten Proteinen, welche einen Anteil von 52 % aller identifizierten Proteine ausmachten, bildeten detoxifizierende und cytoprotektive Proteine mit 10 % Anteil die zweitgrößte Gruppe. Diese Gruppe beinhaltete die Proteine S-formylglutathione hydrolase (ESD), Aryl sulfotransferase (ST1A1), Sulfotransferase 1C1 (ST1C1), Liver carboxylersterase 4 precursor (EST4) und Omega-amidase NIT2 (NIT2) in verminderter Menge. Mit erhöhter Menge wurden Aflatoxin B1 aldehyde reductase member 2 (AKR7A2) und Peroxiredoxin 1 (PRDX1) identifiziert. Mit vermindertem Phosphorylierungsstatus zeigte sich S-formylglutathione hydrolase (ESD) und Catalase (CAT). Für diese Proteine wurde anschließend die Genexpression durch Quantifizierung der spezifischen Transkripte mit Hilfe einer RT-PCR bestimmt und die aktuellste Literatur zusammengefasst. Damit war es möglich, die Veränderung des Proteoms und des Phosphorylierungsstatus in der Cholangiokarzinogenese in der Ratte unter dem Einfluss von TAA zu zeigen und das Rattenmodell als Modell für das Cholangiokarzinom zu bestätigen. Als Fazit aller Ergebnisse dieser Arbeit wird eine Proteinsignatur aus ESD (vermindert), ST1A1 (vermindert), NIT2 (vermindert), PRDX1 (erhöht) und CAT (vermindert phosphoryliert) als diagnostischer und prognostischer Biomarker für die Cholangiokarzinogenese vorgeschlagen.The understanding of underlying molecular and cellular mechanisms during development of an intrahepatic cholangiocarcinoma (iCC) is far from being complete. Connections of various mutations (APC, DCC, OGG1, p53, k-ras, BRAF, TGFßR1, SMAD4) and the involvement of several signalling pathways (PI3K/Akt-, EGFR-, TGF-ß1-, Hippo-, Hedgehog-, Notch-, Wnt-pathway) with iCC have been described. However, it is still unclear whether the underlying mechanism resembles an adenoma-carcinoma sequence and also at which timepoint which specific signalling pathway is disturbed on the proteome level. The aim of this study was the analysis of the proteome and the phospho-proteome of the liver during cholangiocarcinogenesis, induced by thioacetamid (TAA), a water-soluble hepatotoxin. For this purpose, a rat model of male Sprague-Dawley-rats, which were chronically exposed to TAA in the drinking water, was established. Animals were sacrificed after 4, 8, 12 and 16 weeks and assigned to four groups (fibrose; beginning cirrhosis; cirrhosis; cholangiocarcinoma) , based on histological evaluation of liver sections. Further analysis was performed by two-dimensional gel electrophoresis with subsequent mass spectrometry. A total of 38 proteins were identified to be different in protein amount by a factor of more than 1.5, in contrast to the control animal group. Furthermore, 25 proteins were found to differ in the degree of phosphorylation by a factor of more than 1.5, in contrast to the control animal group. The majority of the identified proteins (52 %) could be assigned to a group of proteins, performing metabolic functions; followed by a group of proteins (10 %) related to detoxification and cytoprotection. This second group of proteins comprised S-formylglutathione hydrolase (ESD), Aryl sulfotransferase 1A1 (ST1A1), Sulfotransferase 1C1 (ST1C1), Liver carboxylesterase 4 (EST4) and Omega-amidase NIT2 (NIT2), which were found to be reduced in amounts, compared to the control. The proteins Aflatoxin B1 aldehyde reductase member 2 (AKR7A2) and Peroxiredoxin 1 (PRDX1) also belong to the group of proteins related to detoxification and cytoprotection and were found in increased amounts within this study. S-formylglutathione hydrolase (ESD) as well as Catalase (CAT) were found with reduced degree of phosphorylation. These candidates were further analysed by quantification of the expression of their corresponding genes by mRNA-level determination using RT-PCR. Extensive literature research on the identified candidate proteins is summarized and discussed. In summary, the present study reveals changes in the proteome and the phospho-proteome during TAA-induced cholangiocarcinogenesis in the rat and furthermore confirms the selected rat model being a valid model system to study cholangiocarcinoma. In conclusion, the results of the presented study propose the usage of a protein pattern, consisting of a combination of decreased ESD, ST1A1 and NIT2, increased PRDX1 as well as reduced phosphorylation of CAT instead of a single protein to be used as biomarker for cholangiocarcinogenesis and for prognostic purposes.2017-11-1
