Magnetic resonance imaging in therapy monitoring of liposomal glucocorticosteroids in two animal models of multiple sclerosis in consideration of the size of lesion and lateral ventricle as well as cerebrospinal fluid signal intensity

Die Wirksamkeit liposomaler Glukokortikosteroide wird in zwei Tiermodellen der Multiplen Sklerose anhand einer quantitativen Auswertung T2-gewichteter magnetresonanztomographischer Aufnahmen untersucht. Darüber hinaus werden die Größe der Seitenventrikel und die Liquorsignalintensität der inneren Liquorräume in Beziehung zur ermittelten Krankheitsaktivität gebracht. Die experimentelle Autoimmunenzephalomyelitis der Dark-Agouti- und der Lewisratte zeigt statistisch signifikant verminderte zerebrale Läsionsflächen unter Therapie mit Methylprednisolon bzw. Prednisolon, welche mittels Polyethylenglykol (PEG)-Liposomen verabreicht werden, gegenüber Ratten ohne spezifische Therapie. Gegenüber einer Standardtherapie mit Methylprednisolon können deskriptiv Wirkvorteile beobachtet werden. Für Lewisratten mit Läsionen in der Bildgebung ergeben sich Hinweise, dass vergrößerte Seitenventrikel als rasch erfassbarer Marker der erhöhten Krankheitsaktivität dienen können. Bei Dark-Agouti-Ratten mit Läsionen ist die Liquorsignalintensität hingegen signifikant erhöht. Diese Liquorveränderung korreliert sowohl mit der Läsionslast als auch mit der klinischen Krankheitsausprägung und bietet sich somit als zusätzlicher Krankheitsmarker an.The efficacy of liposomal glucocorticosteroids is examined in two animal models of multiple sclerosis by quantitative evaluation of T2-weighted magnetic resonance imaging. Furthermore the size of lateral ventricles und signal intensity of the inner cerebrospinal fluid space is related to the determined disease activity. Experimental autoimmune encephalomyelitis of Dark Agouti- and Lewisrat is statistically less affected by cumulative cerebral lesion size under therapy with methylprednisolone and prednisolone in polyethyleneglycol (PEG)-modified liposomes compared to rats without specific therapy. Compared to standard therapy consisting of methylprednisolone this effect is noticeable but not significant. For Lewisrats with lesions in imaging there are indications that enlarged lateral ventricles could serve as quick ascertainable marker of higher disease activity. For Dark Agouti rats with lesions however the signal intensity of cerebrospinal fluid is significantly higher. This cerebrospinal fluid alteration correlates with lesion formation as well as clinical symptoms and therefore can function as a marker of disease activity
