Effects of class-3 semaphorins on development and progression of malignant brain tumors

Neuropiline gehören zu den endothelialen Oberflächenrezeptoren und agieren als Rezeptoren für angiogene Faktoren aus der Familie der VEGF-Familie (vascular endothelial growth factor). Daneben spielen sie als Rezeptoren für Faktoren der Axon-Orientierung aus der Klasse-3-Semaphorin-Subfamilie eine Rolle. Kürzlich konnte gezeigt werden, dass Semaphorin-3F (s3f), welches zu den Klasse-3-Semaphorinen zählt, die Tumorgefäßentwicklung hemmt. S3f und andere Klasse-3-Semaphorine wie Semaphorin-3B (s3b) wirken als direkte Hemmstoffe der Tumorzell-Proliferation beim kleinzelligen Bronchialkarzinom. Ferner gibt es erste Hinweise, dass Neuropiline bei der Entwicklung und bei der Progression maligner Hirntumoren eine Rolle spielen. Da Hirntumoren bei Kindern besonders häufig sind und eine vergleichsweise schlechte Prognose aufweisen, sind neue Behandlungsmethoden notwendig, um die schlechten Heilungsaussichten dieser Patienten zu verbessern. In dieser Arbeit konnte gezeigt werden, dass die Überexpression von unterschiedlichen Klasse-3 Semaphorinen in Gliomzellen zu einer Inhibierung des Tumorwachstums führt. Ferner konnten Einflüsse der Semaphorin-Überexpression auf die Migration und die Adhäsion gezeigt werden, die ebenfalls einen Einfluss auf das Tumorwachstum in vivo besitzen können.The neuropilins are endothelial cell surface receptors that function as receptors for the angiogenic factors of the vascular endothelial growth factor (VEGF) family. They also function as receptors for axon guidance factors belonging to the class-3 semaphorin subfamily. Recently, we have shown that semaphorin-3F (s3f), a member of the class-3 semaphorins, functions as an inhibitor of tumor angiogenesis. In addition, s3f, as well as other class-3 semaphorins such as semaphorin-3B (s3b), function as direct inhibitors of tumor cell proliferation in the case of small cells lung carcinoma. There is some preliminary evidence indicating that neuropilins play a role in tumorigenesis and in the progression of malignant brain tumors. Since pediatric brain tumors are characterized by high frequency and poor prognosis, new treatment modalities are imperative to ameliorate the poor prognosis of children suffering from malignant brain tumors. A major aim of this proposal is, therefore, to investigate the antiangiogenic and antitumorigenic potential of class-3 semaphorins in brain tumor cell lines in vitro and in brain tumor models in vivo. We were able to show that overexpression of class-3 semaphorins in glioma cells lead to inhibition of in vivo tumor growth. Additionally the overexpression of class-3 semaphorins lead to changes in migration and adhesion of glioma cells that can influence the in vivo tumor growth, too
