<p><b>(A)</b> RT-QuIC analyses failed to reveal <i>de novo</i> PrP^Sc aggregation when seeded with scFvPOM1 or (scFvPOM1+recPrP_23-230)-treated <i>tg</i>a<i>20</i> COCS homogenates. NBH- and RML6-treated <i>tg</i>a<i>20</i> COCS homogenates were used as negative and positive controls for prion-catalyzed amplification of aggregated PrP, respectively. RT-QuIC reactions were performed in quadruplicates for each sample. <b>(B)</b> <i>tg</i>a<i>20</i> mice inoculated with pooled slice homogenates of scFvPOM1-treated (n = 6 mice) or (scFvPOM1+recPrP_23-230)-treated <i>tg</i>a<i>20</i> COCS (n = 6 mice) stayed healthy for at least 200 days after inoculation. <i>tg</i>a<i>20</i> mice infected at <i>high dose</i> (n = 4 mice) or <i>low dose</i> (n = 5 mice) with RML6 succumbed to prion disease at 55 ± 2 (mean ± s.d., <i>high dose</i>) and 95 ± 5 (<i>high dose</i>) days, respectively.
