The dopamine D2 receptor (D2R) is a member of the GPCR superfamily, and is inextricably linked to a number of important disease states, including schizophrenia and Parkinson’s disease. This thesis focuses on the design and synthesis of novel compounds to probe emerging concepts in D2R structure and function, including allosterism, dimerization, biased agonism, bitopic ligands and bivalent ligands. Firstly, a recent report highlighted SB269652 as the first drug-like allosteric antagonist of the D2R. However SB269652 shares structural homology with a number of orthosteric D2R antagonists and partial agonists.
