The increasing incidence of infections caused by Gram-negative ‘superbugs’ is emerging as one of the most significant threats confronting public health in the modern era. This crisis has been compounded by the decline in antimicrobial drug discovery research over the last 30 years that has resulted in a lack of novel antimicrobial agents in late-stage clinical development. It is against this backdrop that polymyxins, first introduced in the 1950s, have emerged as last-line therapeutic agents for the treatment of recalcitrant infections caused by Gram-negative pathogens, such as Acinetobacter baumannii.
