TLC analysis of cell wall extracts from different <i>C</i>. <i>diphtheriae</i> strains.

<p>(A) Methyl esters of corynomycolic acids (CMAMES) extracted using petroleum ether:acetone (95:5 v/v) from either total lipids (representing trehalose-bound corynomycolates) or delipidated cells (representing cell wall-bound corynomycolates) were separated by TLC and visualized by staining with phosphomolybdic acid (MPA) and charring. (B) TLC analysis of total lipid extracts from <i>Corynebacterium</i> strains using chloroform:methanol:water (60:16:2 v/v/v). Fractions are visualised by charring with alpha naphtol. TDM: trehalose dimycolate; <i>C</i>. <i>glutamicum</i> wild type was used as positive and the <i>pks13</i> mutant <i>Cg-Δpks</i> as negative control. Results for DSM43989 are shown for untransformed strain (wt), transformed with the empty vector pZ8-1 (ev) and complemented by transformation with pZ8-1_<i>B178_03333</i> (ov).
