Cross-presentation of apoptotic cell associated antigen by immature dendritic cells down-regulates IL-7R on CD8 T cells.

<p>2 x10⁶ purified CD45.1+ OT-I T cells were transferred into CD45.2+ mice. One day after the transfer, recipients were challenged with 2 x 10⁶ immature dendritic cells loaded with Apo-BSA, or 2 x 10⁶ immature dendritic cells loaded with Apo-OVA, or 2 x 10⁶ LPS matured dendritic cells loaded with Apo-OVA (n = 5 for each group). At day 7 after challenge, spleen cells from each group were pooled and the transferred OT-I T cells were recovered using anti-CD45.1 biotin beads. RNA from the pooled samples was isolated and reverse-transcribed to cDNA. Relative levels of mRNAs of indicated genes were determined by real-time PCR. Similar results were obtained from two experiments.
