A mutation in a putative nuclear localization signal impairs Acss2 translocation.

<p>(A) Subcellular localization of ectopic V5-tagged wild-type (WT) or cytosol-restricted mutant (CYT) Acss2 in HT1080 cells under basal and stress conditions as detected by immunofluorescence and merging with Hoechst-stained cells to detect nuclei. (B) Subcellular fractionation of stably transformed HT1080 cells with knockdown of endogenous Acss2 and rescue with ectopic WT or CYT mutant Acss2 without or with an SV40 nuclear localization signal fused to the amino terminus. Studies were performed under hypoxia, low glucose, or acetate exposure for the indicated periods.
