Osteoporosis is a common disease, which is caused by both genetic and environmental factors. Osteoporosis increases the risk of fragility fractures, reduces quality of life, and raises mortality rates in affected individuals. Despite years of extensive genetic research, only a few clinically significant underlying genes have been identified. These have been discovered by linkage analysis of families with extreme bone phenotypes. The numerous gene polymorphisms identified by genome-wide association studies have only modest effects on bone mineral density.
