Classic Ehlers-Danlos syndrome is a heritable connective tissue disorder, characterized by fragile and hyperextensible skin, and joint hypermobility. 90% of patients harboured a mutation in the COL5A1 gene, which encode the pro α1(V) chain of the collagen V, that could assemble with the pro α2(V) chain to form the predominant isoform of collagen V, the heterotrimer α1(V) ₂ α2(V).
